血管内皮生长因子(Vascular endothelial growth factor,VEGF)是血管内皮细胞特异性的肝素结合生长因子(heparin-binding growth factor),可在体内诱导血管新生。VEGF有六种不同亚型,包括VEGF-A,-B,-C,-D,及-E;其分子量从35至44kDa不等,每个亚型特异性地与三个“血管内皮生长因子受体”(VEGFR-1,-2,及-3)的特定组合相结合。VEGF是高度保守的同源二聚体糖蛋白。二条分子量各为24kDa的单链以二硫键组成二聚体。VEGF分解的单体无活性,去除N 2糖基对生物效应无影响,但可能在细胞分泌中起作用。由于mRNA不同的剪切方式,产生出分别VEGF121、VEGF145、VEGF165、VEGF185、VEGF206等至少5种蛋白形式,其中VEGF121、VEGF145、VEGF165是分泌型可溶性蛋白,能直接作用于血管内皮细胞促进血管内皮细胞增殖,增加血管通透性。

溶解:提供的人血管内皮生长因子,CHO表达(VEGF CHO)产品为过滤灭菌的白色冻干粉末;该冻干粉末是从含人血管内皮生长因子,CHO表达(VEGF CHO)的磷酸盐缓冲盐水(pH 7.4)中冻干产生的。我们建议加无菌的18MΩ-cm水以配制工作储存液到不低于100ug/ml,后继实验过程中可根据实验要求进一步稀释。
保存:人血管内皮生长因子,CHO表达(VEGF CHO)在室温可保存3周,或置于-18℃冻存用于长期保存。重新溶解后可以在4℃冷藏保存至少2-7天;如果想延长保存时间,建议分装(加载体蛋白如0.1% HSA或 BSA)后置于-18℃冻存。请避免反复冻融。

Introduction
Vascular endothelial growth factor is an important signaling protein involved in both vasculogenesis and angiogenesis. As its name implies, VEGF activity has been mostly studied on cells of the vascular endothelium, although it does have effects on a number of other cell types (e.g. stimulation monocyte/ macrophagemigration, neurons, cancer cells, kidney epithelial cells ). VEGF mediates increased vascular permeability, induces angiogenesis, vasculogenesis and endothelial cell growth, promotes cell migration, and inhibits apoptosis. In vitro, VEGF has been shown to stimulate endothelial cell mitogenesisand cell migration. VEGF is also a vasodilator and increases microvascular permeability and was originally referred to as vascular permeability factor.
Elevated levels of this protein are linked to POEMS syndrome, also known as Crow-Fukase syndrome. Mutations in this gene have been associated with proliferative and nonproliferative diabetic retinopathy.
Description
Vascular Endothelial Growth Factor Human Recombinant produced in CHO cells is a double, glycosylated, polypeptide chain containing 165 amino acids and migrates as 44 kDa in SDS-PAGE under non-reducing conditions.
The VEGF is purified by proprietary chromatographic techniques.

Sterile Filtered White lyophilized (freeze-dried) powder.
Formulation
The protein was lyophilized from a Phosphate- Buffered Saline, pH 7.4.
Solubility (溶解性)
It is recommended to reconstitute the lyophilized Vascular Endothelial Growth Factor Human in sterile 18MΩ-cm H2O not less than 100µg/ml, which can then be further diluted to other aqueous solutions.
Stability (稳定性)
Lyophilized Vascular Endothelial Growth Factor Human although stable at room temperature for 3 weeks, should be stored desiccated below -18°C. Upon reconstitution VEGF Human Recombinant should be stored at 4°C between 2-7 days and for future use below -18°C.
For long term storage it is recommended to add a carrier protein (0.1% HSA or BSA).
Please prevent freeze-thaw cycles.

Greater than 97.0% as determined by:
(a) Analysis by RP-HPLC.
(b) Analysis by SDS-PAGE.
Biological Activity (生物活性)
Determined by its ability to stimulate 3H-Thymidine incorporation in human ublical vein endothelial cells, the ED50 for this effect was found to be 2-6ng/ml, corresponding to a Specific Activity of 166,667-500,000IU/mg.
