干扰素(interferon,IFN)是细胞对相关刺激反应时所产生的细胞信号蛋白质,是细胞因子超家族中一个糖蛋白类同源细胞因子家族,称为IFN家族。已鉴定出该家族中的3个主要成员:IFN-α、IFN-β和IFN-γ。其中干扰素-γ(Interferon-gamma,IFN-γ)是一种具有抗病毒、抗肿瘤和免疫调节作用的细胞因子,主要由活化的T细胞和NK细胞产生,在机体免疫系统中发挥重要作用。IFN-γ是一类分泌型糖蛋白,由143个氨基酸组成,相对分子量是16924。而重组人干扰素-γ相对分子量在14000-16000之间,这是由于重组IFN-γ在C末端有氨基酸丢失现象。
人干扰素-γ重组蛋白是在大肠杆菌中重组表达的,是一条单链、非糖基化的多肽链,包含有144个氨基酸,分子量为16879Da。该人干扰素-γ重组蛋白最终由标准的色谱技术纯化出。

Recombinant Human Interferon-gamma 【IFN g】重组人干扰素-γ细胞因子实验须知:
溶解:提供的人干扰素-γ(IFN g)产品为过滤灭菌的白色冻干粉末;该冻干粉末是从含1mg/ml人干扰素-γ(IFN g)的10mM磷酸钠缓冲液(pH 7.4±0.1)中经进一步透析并冻干产生的。我们建议加无菌的18MΩ-cm水以配制工作储存液到100ug/ml,后继实验过程中可根据实验要求进一步稀释。
保存:人干扰素-γ(IFN g)冻干粉末在室温可保存3周,或置于-18℃冻存用于长期保存。重新溶解后可以在4℃冷藏保存至少2-7天;如果想延长保存时间,建议分装(加载体蛋白如0.1% HSA或 BSA)后置于-18℃冻存。请避免反复冻融。
蛋白含量:蛋白质定量是由两个独立的方法
1。紫外光谱在280海里使用的吸墨性价值0.640的消光系数0.1%(1毫克/毫升)的解决方案。这个值是由电脑计算基因蛋白质序列的计算机分析程序(IntelliGenetics)。
2。产物的分析,使用校准IFN-g作为参考标准的解决方案。

Introduction
IFN-gamma produced by lymphocytes activated by specific antigens or mitogens.
IFN-gamma, in addition to having antiviral activity, has important immunoregulatory functions, it is a potent activator of macrophages, and has antiproliferative effects on transformed cells and it can potentiate the antiviral and antitumor effects of the type I interferons.
Description
Interferon-gamma Human Recombinant produced in E.Coli is a single, non-glycosylated, polypeptide chain containing 144 amino acids and having a molecular mass of 17kDa.
The IFN-gamma is purified by proprietary chromatographic techniques.

Escherichia Coli.
Physical Appearance
Sterile Filtered White lyophilized (freeze-dried) powder.
Formulation
The protein was lyophilized from a 0.2µm filtered concentrated solution in PBS pH 7.4.
Solubility
It is recommended to reconstitute the lyophilized Interferon-gamma in sterile 18MΩ-cm H2O not less than 100µg/ml, which can then be further diluted to other aqueous solutions.
Stability
Lyophilized Interferon gamma although stable at room temperature for 3 weeks, should be stored desiccated below -18°C. Upon reconstitution IFN-gamma should be stored at 4°C between 2-7 days and for future use below -18°C.
For long term storage it is recommended to add a carrier protein (0.1% HSA or BSA).
Please prevent freeze-thaw cycles.
Purity
Greater than 98.0% as determined by:
(a) Analysis by RP-HPLC.
(b) Analysis by SDS-PAGE.

Biological Activity
The specific activity as determined in a viral resistance assay is < 0.05 ng/ml, corresponding to a specific activity of 2.0 x 10,000,000 IU/mg.
Protein content
Protein quantitation was carried out by two independent methods:
1. UV spectroscopy at 280 nm using the absorbency value of 0.640 as the extinction coefficient for a 0.1% (1mg/ml) solution. This value is calculated by the PC GENE computer analysis program of protein sequences (IntelliGenetics).
2. Analysis by RP-HPLC, using a calibrated solution of IFN-g as a Reference Standard.
